この技術では、ごく僅かな遺伝子（DNA）を含む溶液から、端数十から数千塩基対の任意のDNA領域を高速かつ効率的に大々的に増殖させます。細胞が分裂する際、二本鎖のらせん形状を描くDNAは解けて一本ずつに分かれ、それぞれが新たなDNAの鋳型となる過程が酵素によって進行されます。PCRとは、このDNA複製の反応を試験管内で再現し、繰り返し行う手法です（この作業だけで1時間程度でDNA総量を1千万倍にすることも可能です）。
